- It is not possible to determine the concentration of a solution that has a higher y-value than the highest y-value of the standard curve. This is because the plotted data are found along a certain range which if from 0.00 to 0.45 of the line of best fit that has been plotted. Therefore, values outside these range would not give an accurate protein concentration.

PART 2

- The experiment should be repeated at least 3 times to obtain a more reliable answer. This is done my calculating the average of each of the repeated experiments. Also, wider range of known protein concentration should be used to plot points on the standard curve to ensure unknown concentration of protein falls on a wider range of protein concentration.

- Result does not support hypothesis. Leaves that are grown in sun has a higher concentration of protein compared to those grown in shade.

- The possible sources of errors are, error in preparing the modified solution when carrying out dilution and volume of sample use to fill into the cuvette is too little or too full. The base and exterior part of the cuvette was not wiped dry, therefore condensation may interfere dually. Also, the non-clear section of the cuvette placed facing the source of light in spectrophotometer may cause deflection of light.

- Presence of organelles in sample may affect the quantity of light passing through the sample which may result in light scattering than passing through sample. Furthermore, surrounding temperature may not be suitable to carry out experiment as it may cause denaturation of enzyme which may influence the protein concentration of sample, thus resulting in negative result. Also, the change in pH value may affect the enzyme activity of the solution causing enzyme to loose it’s tertiary structure, thus causing a negative result in the experiment.

- Light is the only factor taken into consideration as a significant effect on absorbance accuracy. Furthermore, the spectrophotometer only shows one reading scale for each protein samples.
