- Make up 5 test tubes according to the following table

- Record the pH for each of the five test tubes using pH paper

- Add 2 mL deionized water, 8 drops of catechol, and 8 drops of the potato extract into all five test tube

- Mix gently and place the tubes into a 37 degrees Celsius water bath.

- After 15 minutes, examine each tube for color changes and compare the intensity of color. The development of color is visual evidence of the oxidation process catalyzed by the enzyme. All colors should be rated on a scale of 1–10 with 10 being the darkest. Record the data in Table 3 of the lab worksheet

Experiment 5: Effect of Temperature on the Rate of Enzyme Reaction

- Place 10 drops of the potato extract and 1.5 mL deionized water into the five test tubes

- Place 10 drops of 0.01 M catechol solution (substrate) and 2 mL deionized water into another set of five test tubes

- Create a 0 degrees Celsius water bath in a beaker using water, ice, and salt

- Create a 10 degrees Celsius water bath in a beaker using water and ice

- Create a room-temperature water bath in a beaker using tap water

- Use the 37 degrees Celsius water bath in the back of the lab

- Heat a glass beaker of deionized water on the hot plate to 70 degrees Celsius

- Monitor and maintain temperatures carefully with the thermometers provided

- Place one of each tube (one potato extract and one substrate) into each of the five water baths for 10 minutes

- Quickly combine the contents of each set of tubes (one enzyme for every temperature substrate) and return them immediately to the correct water bath for an additional 15 minutes

- After 15 minutes, examine each tube for color changes and compare the intensity of color. The development of color is visual evidence of the oxidation process catalyzed by the enzyme. Record the data in Table 5 of the lab worksheet

- Place all tubes into the water bath at 37 oC for 10 minutes to see if the color intensifies. If it does, then the enzyme was just slowed down by the temperature change. If no color appears, then the enzyme was permanently damaged by the temperature. Record the data in Table 5 of the lab worksheet

- Prepare how the results will be presented to the class. Include why different incubation temperatures were tried, what should have been seen, and what was seen
